CoregHydrido complexes, especially of the platinum group metals, have a pivotal role in homogeneous catalysis 1 ; . They appear as intermediates in many catalytic cycles, including hydroformylation, asymmetric hydrogenation, hydroxycarboxylation, and oligomerisation of olefins. In order to perform highly selective hydrido-mediated catalytic reactions it is essential that ligands are present which allow both fine-tuning of the M-H bond reactivity and proper discrimination of incoming substrates, with thc latter being especially important for enantioselectivity.A major limitation of presently available platinum metal hydride catalysts concerns the freely accessible reactive metal centre. With such species, there is no easy way to discriminate between incoming substrates of different shape. A difficult challenge to be overcome during the development of the next generation of hydride catalysts is to locate the platinum metal hydrides inside well-defined molecular pockets. Such structures are expected to possess valuable properties, in particular with respect to shape-selectivereactions, that is, where the cavity functions as a molecular recognition unit by which to channel the substrate to the catalytic site in a preferred orientation, see Scheme I. Possible synthons for the construction of appropriate molecular cavities are the calix [4]arenes, but there has been little success in combining them with reactive metallo-fragments. Calix[4]arenes L ; constitute a class of versatile building blocks that have recently become very useful in the general area of molecular engineering 2 ; . They are cone-shaped macrocyclic compounds comprising four phenolic units linked together in a circular array by methylene bridges connected at the ortho positions of the phenolic rings. An essential feature is the presence of phenoxy rings oriented in the same direction, so that, in principle, tethering of functional groups either to the phenolic oxygen atoms or to the pcarbon atoms generates well-organised cavities which may provide attractive binding zones for incoming ionic or neutral species. Thus, for instance, the p-derivatised calixarene L', see Scheme 11, in which the cone shape is retained, displays remarkable complexing properties towards actinides, with a binding affinity being some 200-fold better than that of the monomeric counterparts, namely the 0, O-chelators, formula R, P 0 ; CH2C O ; NRR 3 ; . Background: The goal of this project is to investigate whether RV-PCR can be used to generate profiles from LCN samples 100 picograms of template DNA ; using ABI's IdentifilerTM system. Four reduced PCR volumes 15L, 10L, 5L, and 2.5L ; will be evaluated to determine the most reliable reaction volume based upon the results of sensitivity studies, reaction volume performance under concentrated EDTA conditions with and without addition of mgCl2 during re-hydration ; , reproducibility studies, and the ability to interpret twocomponent mixtures. The performance of the optimal reduced reaction volume will then be evaluated using a simulated LCN forensic DNA case. Finally, a validated RV-PCR protocol for use in routine forensic casework involving LCN samples at the SCLFS will be developed. Methods: Cells will be collected from two male donors somatic and spermatic ; and one female somatic only ; donor. DNA will be extracted using a standard organic protocol with DNA quantification using ABI's real-time PCR QuantifilerTM system. STR amplification will be performed using ABI's IdentifilerTM kits with the PCR products separated by capillary gel electrophoresis on an ABI 3100 Genetic Analyzer and genotyped using GeneMapperIDTM software. Sensitivity studies will be performed with DNA template ranges of 1500pg to 100pg for the full reaction volume RV ; , 900pg to 60pg for the 15L RV, 600pg to 40pg for the 10L RV, 300pg to 20pg for the 5L RV, and 150pg to 10pg for the 2.5L RV. The dehydration rehydration study will use two sets of four DNA extraction recovery volumes 50L, 25L, 10L, and 5L ; at a series of DNA template levels 500pg, 250pg, 100pg, and 10pg ; . Set #1 will be reconstituted with sterile water and Set #2 with sterile water plus an equal molar concentration of mgCl2 equivalent to the amount of EDTA in the initial recovery extract. Two LCN mixture studies will be performed somatic: somatic and spermatozoa: somatic ; at a 20: 1, 10: and 1: ratio for each component. Finally, two mock-LCN sexual assault biological evidence samples will be prepared at LCN levels. Somatic- and spermatic-derived DNA will be separated using a modified differential extraction protocol, and RV-PCR will be performed under optimal conditions Results: Complete STR profiles have been developed from all reaction volumes. Using a large number of plant microarray analyses to help pinpoint the mechanisms of gene regulation is limited by the range of tools currently available. We have developed ModuleFinder to identify sets of genes and treatments that in our hands contain more biologically related functions for analysis of the mechanisms behind co-expression in non-linear-related sets. We then developed CoReg to link the clustering tree of expression-based relationships in these gene sets with frequency tables of promoter elements. These sets of promoter elements represent putative CAREs for sets of genes, and can then be tested experimentally. We consider these tools, both built on an open source software product, provide a valuable alternative tool to those widely available for the prioritisation of promoter elements for experimental analysis and lotensin.
